-Sometimes ending the CE run early makes sense when my data collection is complete as long as there are size standard peaks on each side of data. However, this results in yellow icons (A) next to sample names indicating poor size calling and scores (B) due to a truncated actual size standard compared to expected manufacturer size standard length (C).
-To improve the score and accuracy of size calling, you can create a modified size template with the following instructions:
- Go to Tools -> Size Template Editor
-Upon opening, you'll see top electropherogram shows expected Size Standard pattern that goes to Size 120.0 for this example. The bottom electropherogram shows truncated actual Size Standard that goes to Size 80.0.
2. Go to File -> Save As New Size Standard
3. Rename file with descriptive name and click OK
4. Right-click and Delete Size values that are not included in your data set (either from the electropherogram or table).
Note: in this example we delete 110.0, and 120.0 because the CE run stopped after Size 80.0
-With the new edits, now the actual size standard matches the expected size standard. When you close this screen, the size calling updates automatically.
-Now icons to left of sample in Allele Call folder are green and the size calibration chart shows improved scores, which means the size calling of your sample peaks is more accurate.
